Eli Lilly and Company is significantly expanding its footprint in Indiana, announcing an additional $4.5 billion investment that brings its total commitment to the state to over $21 billion since 2020.

This latest infusion of capital, highlighted by President Donald Trump on Truth Social following a Wednesday press release, will bolster two locations in Lebanon: the future Lilly Lebanon API facility for active pharmaceutical ingredients and Lilly Lebanon Advanced Therapies.

The latter officially opened on Wednesday, marking a milestone as the company’s first facility dedicated exclusively to the manufacturing of genetic medicines.

The company described Lebanon as the cornerstone of its domestic manufacturing buildout. In 2024, Lilly announced plans to make the weight management and type 2 diabetes medications, Zepbound and Mounjaro, at the Lebanon API site.

The investment announced on Wednesday further expands the commitment and includes the planned production of the weight loss pill Foundayo and retatrutide, which is in late-stage development for obesity and cardiometabolic disease, Lilly explained.

“Lilly’s legacy of firsts in Indiana continues today–and the best measure of that legacy is what we do next,” said Lilly chair and CEO, David Ricks.

“From genetic medicines that could one day prevent disease at its source, to Foundayo, a pill making weight loss treatment accessible to millions, we are not just discovering the medicines of the future–we are building the world’s most advanced plans to make them,” he continued.

Ricks went on to say that the Lebanon API site, scheduled to open in 2027, would be the largest API production site in U.S. history.

“This expansion reflects the strength of a long-standing partnership between Lilly and the state of Indiana — one that continues to deliver real results for Hoosiers,” said Governor Mike Braun (R-Ind.).

Bolstering its claim as a cornerstone of the regional economy, Lilly cited an upcoming report from Indiana University’s Kelley School of Business and the Indiana Business Research Center, which estimates that the company accounts for a staggering 70% of Indiana’s pharmaceutical GDP.

Beyond state lines, the company credited a favorable domestic policy environment for its broader national expansion. This momentum is set to continue through the end of the year, as Lilly plans to break ground on several new manufacturing sites across the United States to further strengthen its domestic supply chain.

“Lilly’s U.S. capital expansion commitments since 2020 total more than $50 billion, investments made possible thanks to policies that promote domestic manufacturing,” the release said.
